Reaction Principle

Some bacteria decompose glucose in glucose peptone water medium to generate pyruvic acid. Pyruvic acid undergoes condensation and decarboxylation to form acetoin. Under strong alkaline conditions, acetoin is oxidized by oxygen in the air to diacetyl. Diacetyl reacts with guanidyl groups in peptone to produce a red compound, which is defined as a positive V-P reaction.
