Operating Procedures
- Inoculate pure bacterial culture into 1 mL tryptophan broth, tryptone water or other suitable medium, incubate at 36 ± 1°C for 24 ± 2 h.
- Add 2–4 drops of the reagent and mix thoroughly.
- Observe the result immediately, or leave at room temperature for 5–10 minutes before observation.
Reaction Principle
Certain bacteria can decompose tryptophan in peptone to produce indole. Indole can be detected via color reaction: indole combines with p-dimethylaminobenzaldehyde to form rosindole, a red-colored compound.
Result Interpretation
- Positive: A distinct red ring appears on the upper layer of the medium.
- Negative: The solution turns yellow, orange or brown without red ring formation.
